Amphotericin B

Amphotericin B is a polyene antibiotic.
Polyene antibiotics are produced from a Streptomyces species. They interact with fungal cell membrane, sterols, primarily ergosterol, which causes increased permeability of cell wall that leads to cell lysis.

Mechanism of Action:

The mechanism of action of amphotericin B is concentration dependent. Amphotericin is polyene with larger molecule, and unlike natamycin (Natamycin is a smaller molecule polyene antibiotic,read about Natamycin) works by creating ‘pores’ in the cell wall, allowing ions like potassium to leak through the cell wall. This causes imbalance in the osmotic gradient and subsequent cell death.

Pharmacokinetics:

Subconjunctival injection of amphotericin is not recommended because of severe toxicity
